OASIS Implant a ground-breaking system that is capable of performing simultaneous all-optical recording (e.g. Calcium imaging) and manipulation (e.g. optogenetic stimulation) in the deep brain of a freely behaving animal to observe neural circuit activity with high cellular resolution. Compatible with high-sensitivity research-grade cameras, OASIS Implant enables researchers to acquire high quality deep-brain images with low noise and high linearity, enabling quantitative data analysis. In addition, with multiple illumination ports and multiple filter slots, OASIS Implant can be reconfigured by end users to precisely fit their specific experimental needs. Equipped with both wide-field and patterned illumination capabilities (when combined with Mightex's market-leading Polygon400), OASIS Implant allows scientists to simultaneously perform Calcium imaging and targeted optogenetic stimulation with pin-point accuracy to stimulate individual neurons (but without unwanted stimulation of the neighbouring neurons). Moreover, it is also capable of multi-color imaging and optogenetic stimulation. Another key benefit of the OASIS Implant system for behavioral neuroscience is its super light-weight head mount. Since both deep-brain imaging and stimulation are delivered through a very thin imaging fiber, the overall weight of the head-mounted fixture is as little as 0.7g (compared to approximately 2~3g with a head-mounted camera), which minimizes stress experienced by the animal, and allows for the duration of the experiments to be extended. All of these features combined make the OASIS Implant an ideal research tool for in vivo experiments.

The OASIS Implant provides single-cell and population resolution imaging for a wide-range of experimental setups. With a standard c-mount camera port, OASIS Implant accepts any cameras, including research-grade scientific cameras which enable scientists to capture high-quality deep-brain images and videos. Furthermore, the OASIS Implant is compatible with multiple light sources to image different imaging indicators and fluorophores. Thus, the OASIS Implant will enable scientists to acquire more detailed and more accurate data for not only qualitative, but also quantitative analysis.

The Imaging Fiber Probe (IFP) consists of thousands of individual mini-fibers or pixels, and each of these pixels can be independently addressed with Miughtex's market-leading Polygon400 patterned illuminator. Therefore, the OASIS Implant can be used to deliver light to deep brain regions with sub-cellular resolution, and this opens the door for scientists to conduct experiments, such as deep-brain sub-cellular optogenetic stimulation or targeted calcium imaging, in a freely-moving animal. In addition, traditional wide-field illumination can easily be supported by sending light through all the individual pixels in the IFP. The OASIS Implant platform has two illumination ports, each capable of taking either a wide-field or a patterned illuminator with single or multiple wavelengths/colors.

fiber, or multiple groups of fibers for high cellular stimulation resolution in vivo.
The OASIS Implant enables imaging and stimulation in a freely-moving animal. This is due, in part, to the light-weight headmount (<0.7g) and the system being external from the animal. These requirements will help extend the length of experiments and reduce stress for imaging and stimulation Experiments.font>

With included software, users can acquire a continuous stream of data from within the deep brain to identify and define neurons, and then extract intensity traces for each cell. Calcium traces can also be synchronized and time-stamped with other behavioural equipment (cameras, behavioural equipment etc.) to generate precise and comprehensive data for analysis.

As shown in the schematic diagram below, a typical OASIS Implant system contains the following parts:
(1) At the heart of the system, an OASIS Implant platform that connects all the parts together; (2) An Imaging Fiber Probe (IFP, also called "Fiber Bundle") that consists of thousands of mini-fibers, each of which can be independently addressed. OASIS Implant supports both uniform wide-field illumination and patterned illumination when used with a Polygon400 patterned illuminator. The versatility of the OASIS Implant allows researcher to use both illumination methods simultaneously. One example is to use patterned illumination to optogenetically stimulate selected neurons while observing fluorescence images with the wide-field illumination. Another example is to use structured illumination from the patterned illuminator to suppress background during fluorescence imaging.
System Configurations
One Platform, Any Experiment
The OASIS implant is a generic all-optical imaging and stimulation platform designed for diverse in vivo neuroscience experiments. With such a platform, one can easily reconfigure the OASIS implant system to suit his/her current and future experimental needs, such as Calcium imaging and deep-brain targeted optogenetic stimulatioin etc. This can be achieved by adding/removing light sources, dichroics, and cameras etc. to/from the OASIS Implant platform, providing users with a futureproof solution capable of addressing any new applications. Listed below are some typical OASIS Implant application examples and corresponding system configurations.
Application Example-1: Calcium Imaging
This is the OASIS Implant base configuration. With this configuration, researchers will be able to image a single calcium indicator (e.g. GCaMP) in the deep-brain of a freely-moving animal using a single wide-field LED. This configuration is an excellent starting point for many researchers, since the base configuration can be easily expanded into any of the following setups.
Application Example-2: Calcium Imaging and Targeted Optogenetics
This OASIS implant configuration will enable researchers to image a single calcium indicator (e.g. GCaMP)/fluorophore (e.g. GFP) or perform targeted optogenetic stimulation (e.g. ChR2) in the deep brain of a freely-moving animal by adding our Polygon400 patterned illuminator to the base configuration (blue).
Application Example-3: Dual-Colour Imaging
This OASIS implant configuration will enable researchers to image two different colour calcium indicators (e.g. GCaMP and RCaMP) in the deep-brain of a freely-moving animal using two wide-field illumination sources. From the base configuration (blue), this configuration adds a second LED source.
Application Example-4: Simultaneous Calcium Imaging and Targeted Optogenetics
This OASIS implant configuration will enable researchers to simultaneously image a calcium indicator (e.g. GCaMP)/fluorophore (e.g. GFP) and perform targeted optogenetics (red-shifted ChR2) in the deep-brain of a freely-moving animal. From the base configuration (blue), this configuration adds a Polygon400 patterned illuminator.
Application Example-5: Advanced All-Optical Imaging and Targeted Stimulation System
This OASIS implant configuration is the complete package and will enable researchers to perform every capability listed above and more. The system includes two Polygon400 patterned illuminators. Researchers will be able to perform the most advanced experiments, such as dual-colour targeted optogenetics (e.g. ChR2 and NpHr), dual-colour imaging, or simultaneous imaging and targeted optogenetics in the deep-brain of a freely-moving animal.
